Expanding platelet transfusion therapy in Ireland
HRB Applied Partnership - Cold-Stored Platelets
Background
Introduction of Cold-Stored Platelets (CSP) is complicated by the lack of CSP-specific quality control measures
Aim
- Characterisation of cold-stored platelet production methodologies and functionality to expand platelet transfusion therapy in Ireland
- Building on the expert knowledge and existing collaborative experience between the Royal College of Surgeons in Ireland (RCSI; Researcher) and the Irish Blood Transfusion Service (IBTS; Knowledge User), this initiative will address these needs as follows:
- Work Package 1: Minimise the build-up of inflammatory mediators in CSP which could lead to adverse transfusion reactions.
- Work Package 2: Investigate the role of platelet additive solutions on platelet phenotype and functionality.
- Work Package 3: Novel approaches to identify platelet signalling pathways associated with declining platelet functionality during cold storage by employing RNA sequencing and a comprehensive set of platelet function tests.
- Work Package 4: Use specific additives to counteract cold-induced platelet storage lesions.
Impact
The knowledge generated by this research will provide an evidence base to:
- Facilitate local decision-making at the IBTS regarding the introduction of CSP as a new blood component
- Support the development of European-wide CSP-specific quality control parameters, optimal storage conditions
- Delineating needs and suitable approaches for further research into CSP.
TU-Dublin Business Partnership Scheme - Characterisation of Platelet-Derived Extracellular Vesicles
Background
The factors that influence the quality of blood components are incompletely understood, leading to waste and sub-optimal clinical outcomes. "What are the molecular changes that occur in platelet-pack associated extracellular vesicles, and how can these be exploited to improve platelet quality and reduce waste?"
Aim
- Define the physical, biochemical and functional changes that occur to platelet-derived extracellular vesicles (PEV) during the preparation, processing and storage of donated blood.
- Develop models to more accurately predict a donor-associated lifespan of blood products supporting platelet component preservation.
